利用 R T P C R 技术, 我们检测了 P19 细胞体外神经元分化过程中神经钙粘着蛋白( Ncadherin) 的表达模式。结果显示, 该基因在上述过程中存在上调和下调过程, 与体内中枢神经系统发育过程的表达模式十分相近。在此基础上, 我们将神经钙粘着蛋白基因c D N A 全长转入 P19 细胞, 通过药物筛选, 得到了稳定表达神经钙粘着蛋白的细胞株。转基因细胞株不经 R A 诱导, 就能自发向神经元方向分化, 并表达神经元特异的分子标记物 N F160 。以上结果提示: 神经钙粘着蛋白可引起 P19 细胞在体外自发向神经方向分化, 在 P19 神经元的分化过程中起重要作用。
Using RT-PCR, we examined the expression of N-cadherin in P19 cells in vitro during neuronal differentiation. The results showed that there was up-regulation and down-regulation of this gene in the above process, which was similar to the expression pattern of CNS development in vivo. On this basis, we transferred the full-length cD N A of the neural cadherin gene into P19 cells and obtained the cell strain stably expressing the neural cadherin through drug screening. Transgenic cell lines can spontaneously differentiate into neurons without R A induction and express neuron-specific molecular marker N F160. These results suggest that: neuronal cadherin can cause P19 cells in vitro to spontaneous neural differentiation, P19 neurons play an important role in the process of differentiation.
