目的 观察甘糖酯对大鼠尿激酶的诱导作用 ,从基因水平上探讨甘糖酯的抗栓机制。方法 采用发色底物法检测甘糖酯对大鼠血浆中尿激酶活性的影响 ,以半定量RT PCR法检测大鼠尿激酶基因表达。结果 大鼠经灌喂甘糖酯后血液中尿激酶活性和mRNA表达水平均高于对照组 ,且尿激酶活性和mRNA表达水平的升高同喂药剂量正相关 ,在 2 5~ 10 0mg·kg-1的剂量范围内随剂量增大而升高。结论 甘糖酯可以在mRNA水平上诱导大鼠尿激酶的表达 ,或加快大鼠尿激酶mRNA的转录速度或增加mRNA的稳定性 ,激活纤溶系统 ,从而表现出一定的抗栓活性
Objective To investigate the induction of urokinase on rat urokinase and to explore the antithrombotic mechanism of the glyceryl ester at gene level. Methods The chromogenic substrate method was used to detect the effect of glycol ester on the activity of urokinase in rat plasma. The mRNA expression of urokinase in rat was detected by semi-quantitative RT-PCR. Results The levels of urokinase and mRNA in blood were higher than those of the control group, and the increase of urokinase activity and mRNA expression was positively correlated with the dose of feeding, kg-1 dose range with increasing doses. CONCLUSION Glycopeptide can induce the expression of urokinase in rats at mRNA level or accelerate the mRNA transcription of urokinase in rats or increase the stability of mRNA and activate fibrinolytic system and thus show some antithrombotic activity
