通过比较11种大孔树脂对甜茶总黄酮的吸附和解吸效果,筛选出适合甜茶总黄酮分离纯化的树脂,并对其纯化工艺条件进行了探讨。结果表明,FL-3型大孔吸附树脂最适合甜茶总黄酮。最佳吸附条件为:上柱液总黄酮质量浓度控制在2.069 mg/mL~3.104 mg/mL,上样流速2 BV/h;上样液体积控制在4.5 BV以内;最佳洗脱条件为:先用蒸馏水洗去水溶性杂质,再用8.3 BV 40%乙醇洗脱,收集40%乙醇洗脱部分。经FL-3型大孔吸附树脂纯化甜茶总黄酮的纯度可达到26.64%。
By comparing the adsorption and desorption effects of 11 kinds of macroporous resin on the total flavonoids of sweet tea, the resin suitable for separation and purification of the total flavonoids of sweet tea was screened and the purification process conditions were discussed. The results showed that FL-3 macroporous resin is the best for the total flavonoids of sweet tea.
