目的 建立螨类细胞培养方法 ,为从细胞分子水平研究革螨、恙螨作为HFRS媒介提供条件。方法 采用革螨、恙螨的若虫、幼虫进行螨类细胞培养 ,建立螨细胞系。革螨、恙螨若虫、幼虫经消毒、胰酶消化处理后 ,接种于含15 %FBS改良TC199培养基中培养。结果 两种螨细胞 2周后细胞生长良好 ,并形成单层 ,现已传了 4代。细胞形态以梭形为主 ,染色体 2n =6 ,细胞对数期群体倍增时间 2 0 .5 0h。最能适应在TC199培养基上生长。动态观察对氨基酸要求 ,发现谷氨酸、酪氨酸、鸟氨酸 ,胱氨酸和精氨酸需量最大。结论 本结果为研究HFRSV在螨体内增殖、定位提供了条件和方法
OBJECTIVE: To establish a mite cell culture method for the research of leather mite and chigger mite as a medium of HFRS at the cellular and molecular level. Methods Using mites and chiggers nymphs and larvae, mite cells were cultured to establish mite cell lines. Leather mites, chigger nymphs, larvae after disinfection, trypsinization, inoculated in 15% FBS modified TC199 culture medium. Results Two kinds of mite cells grew well after 2 weeks and formed a monolayer, which has been passed for 4 generations. Fusiform cell morphology, chromosome 2n = 6, cell doubling time population doubling time 20.5h. Most adapted to TC199 medium growth. Dynamic observation of amino acid requirements, found that glutamate, tyrosine, ornithine, cystine and arginine demand most. Conclusion The results provide the conditions and methods for studying the proliferation and localization of HFRSV in mites
