目的建立微波辅助萃取-高效液相色谱(MAE-HPLC)法分析金银花药材中槲皮素和木樨草素的含量。方法采用Diamonsil C18柱(150 mm×4.6 mm,5μm),以甲醇-0.2%三氟乙酸(55∶45,V/V)为流动相,流速1.0 mL.min-1,检测波长350 nm。微波辅助萃取条件:2.5 mol.L-1盐酸甲醇溶液10 mL,微波功率为360 W,提取时间为3 min。结果槲皮素在5~100 mg.L-1内线性关系良好(r=0.999 5),平均加样回收率为92.35%,RSD为1.33%(n=3);样品的日内和日间RSD分别为0.44%和1.61%。木樨草素在0.25~5 mg.L-1内线性关系良好(r=0.999 9),平均加样回收率为92.40%,RSD为1.13%;日内和日间RSD为0.79%和2.60%。结论该方法快速、准确,操作便捷,适用于提取分析和测定金银花中槲皮素和木樨草素的含量。
Objective To establish a microwave-assisted extraction-high performance liquid chromatography (MAE-HPLC) method for the determination of quercetin and clodronine in honeysuckle. Methods Diamonsil C18 column (150 mm × 4.6 mm, 5 μm) was used with a flow rate of 1.0 mL · min-1 with methanol-0.2% trifluoroacetic acid (55:45, V / V) Microwave-assisted extraction conditions: 2.5 mol.L-1 methanolic hydrochloric acid solution 10 mL, microwave power of 360 W, extraction time was 3 min. Results The linearity of quercetin in the range of 5-100 mg · L-1 was good (r = 0.999 5), the average recovery was 92.35% and the RSD was 1.33% (n = 3). The intra- and inter-day RSD Respectively 0.44% and 1.61%. The linear correlation was found between 0.25 and 5 mg.L-1 for woodcurcumin (r = 0.999 9), with an average recovery of 92.40% and a RSD of 1.13%. The intraday and interday RSDs were 0.79% and 2.60%, respectively. Conclusion The method is rapid, accurate and easy to operate. It is suitable for the extraction and analysis of quercetin and clodronine in honeysuckle.
