作者应用Percoll对PHA体外刺激的正常人外周血淋巴细胞活化后的淋巴母细胞进行分离,并用碱性磷酸酶-抗碱性磷酸酶(APAAP)免疫组化技术对纯化后细胞的表型和干扰素-γ(IFN-γ)的表达进行了检测。结果表明:淋巴母细胞收获率约为80%,纯度约达90%。通过对纯化后细胞的各种活化抗原(Tac,A1,T9,Ia)和IFN-γ的表达进行检测表明纯化的细胞主要为淋巴母细胞。本实验所建立的淋巴母细胞分离方法具有母细胞得率高,纯度好和方法简便等优点,为研究某些传染病患者外周血中出现的异型淋巴细胞提供了较理想的分离方法。
Percoll was used to separate the lymphoblasts from normal human peripheral blood lymphocytes stimulated by PHA in vitro. The phenotype and interference of the purified cells were detected by alkaline phosphatase-anti-alkaline phosphatase (APAAP) immunohistochemistry The expression of ubiquitin-gamma (IFN-γ) was examined. The results showed that the rate of harvest of lymphoblasts was about 80% and the purity was about 90%. The purified cells were mainly lymphoblasts by the detection of the expression of various activated antigens (Tac, Al, T9, Ia) and IFN-γ of the purified cells. The lymphoblast cell separation method established in this experiment has the advantages of high yield of mother cells, good purity and simple method. It provides an ideal separation method for the study of abnormal lymphocytes in peripheral blood of some infectious diseases.
