目的 :观察雷公藤甲素 (T1 0 )对肿瘤坏死因子 (TNF)、白细胞介素 6 (IL - 6 )在平滑肌细胞 (SMC)、腹腔巨噬细胞(MΦ )的分泌以及在小鼠体内诱生的影响。 方法 :体外主动脉 SMC培养 ,腹腔注射给药及眼球摘除取血 ,细胞毒结晶紫染色法测定 TNF,依赖细胞株 MTT比色法测定 IL - 6。结果 :T1 0 注射浓度 (0~ 0 .375 mg/ kg)与小鼠血清中 TNF,IL - 6水平呈显著负相关 (P<0 .0 1) ;腹腔 MΦ 培养上清中 TNF及 IL- 6分泌与 T1 0 给药浓度 (0~ 1.1μm ol/ L)呈显著负相关 (P<0 .0 1) ;0 .2 75μmol/ L T1 0 对指数增殖期 SMC自分泌及在脂多糖 (L PS)、氧化修饰低密度脂蛋白胆固醇 (ox L DL)刺激下分泌 TNF,IL- 6 ,TNF和 IL- 6的相互诱生亦有明显抑制 (P<0 .0 1)。 结论 :T1 0 可以抑制 TNF,IL- 6的体内外诱生。
Objective: To observe the effects of triptolide (T1 0 ) on the secretion of tumor necrosis factor (TNF) and interleukin 6 (IL-6) in smooth muscle cells (SMC) and peritoneal macrophages (MΦ ) and in mice Health effects. METHODS: In vitro aortic SMC culture, intraperitoneal injection and enucleation were used for blood sampling, cytotoxic crystal violet staining for TNF, and cell-dependent MTT colorimetric assay for IL-6. RESULTS: T1 0 injection concentration (0 to 0.375 mg/kg) was significantly negatively correlated with TNF, IL-6 levels in serum of mice (P < 0.01); TNF and IL- in supernatants of peritoneal MΦ cultures 6 secretion was significantly negatively correlated with T1 0 dose (0-1.1 μmol/L) (P <0. 01); 0. 2 75 μmol / L T1 0 auto-secreted on exponentially proliferating SMC and in lipopolysaccharide ( LPS) and oxidatively modified low density lipoprotein cholesterol (ox LDL) stimulated secretion of TNF, IL-6, TNF and IL-6 were also significantly inhibited (P < 0.01). Conclusion: T1 0 can inhibit the in vitro and in vivo induction of TNF and IL-6.
