目的考察不同质量浓度激素组合对甘西鼠尾草愈伤组织诱导、增殖及分化的影响。方法以甘西鼠尾草叶片为外植体,采用正交试验研究2,4-D、NAA、6-BA、KT对愈伤组织诱导及增殖的影响,并探讨适宜愈伤组织分化的培养条件。结果以诱导率和相对生长速率为指标,6-BA和2,4-D均对甘西鼠尾草细胞脱分化和细胞增殖具有显著影响,愈伤组织诱导的最佳激素组合为2,4-D 0.5 mg/L+NAA 1.0 mg/L+6-BA 0.5 mg/L,增殖的最佳激素组合为2,4-D 0.5 mg/L+6-BA 2.0 mg/L+KT 0.1 mg/L,MS+NAA 0.5 mg/L+6-BA 1.0 mg/L的培养基有利于不定芽的分化,而1/2 MS+IBA 1.0 mg/L的培养基则有利于不定根的分化。结论本实验所建立的组织培养体系,适宜甘西鼠尾草的植株再生。
Objective To investigate the effects of different concentrations of hormone combinations on callus induction, proliferation and differentiation of Salvia miltiorrhiza. Methods The explants were used as explants. The effects of 2,4-D, NAA, 6-BA and KT on the induction and proliferation of callus were studied by orthogonal test and the culture of suitable callus differentiation condition. Results The induction rate and relative growth rate were used as indexes. Both 6-BA and 2,4-D had significant effects on the dendifferentiation and cell proliferation of S. gansuensis cells. The best hormone combination induced by callus was 2,4 -D 0.5 mg / L + NAA 1.0 mg / L + 6-BA 0.5 mg / L, and the best hormone combinations for proliferation were 2,4-D 0.5 mg / L and 6-BA 2.0 mg / L and MS + NAA 0.5 mg / L + 6-BA 1.0 mg / L could promote adventitious bud differentiation, while 1/2 MS + IBA 1.0 mg / L medium was conducive to adventitious root differentiation. Conclusion The tissue culture system established in this experiment is suitable for plant regeneration of Salvia miltiorrhiza.
