目的:合成并表征透明质酸接枝十八烷基(HA-C18)两亲性聚合物。方法:通过十八烷胺的氨基与透明质酸的羧基发生酰化反应,制得HA-C18聚合物,采用FTIR鉴定合成的HA-C18聚合物的化学结构,评价HA-C18聚合物的自聚集行为,测定其临界聚集浓度;以可静脉注射辅料Tween 80和Cremophor EL为参比,评价HA-C18聚合物的溶血性。结果:合成的HA-C18聚合物临界聚集质量浓度10.0 mg.L-1,HA-C18胶束溶血性远远低于Tween 80;Cremophor EL溶血性略低于HA-C18胶束,但差异不明显。结论:HA-C18聚合物是优良的可用于静脉注射的难溶性药物载体材料。
Objective: To synthesize and characterize HA-C18 amphiphilic polymers. Methods: The HA-C18 polymer was obtained by the acylation of the amino group of octadecylamine with the carboxyl group of hyaluronic acid. The chemical structure of HA-C18 polymer was identified by FTIR. Aggregation behavior was measured to determine the critical aggregation concentration. The haemolytic activity of HA-C18 polymer was evaluated by using intravenous adjuvant Tween 80 and Cremophor EL as reference. Results: The critical concentration of HA-C18 polymer was 10.0 mg.L-1, the HA-C18 micellar hemolytic value was much lower than that of Tween 80. The hemolytic activity of Cremophor EL was slightly lower than HA-C18 micelles, but the differences were not significant obvious. Conclusion: HA-C18 polymer is an excellent drug carrier material for intravenous injection.
