目的研究胃腺癌组织中瘦素mRNA表达及其细胞定位,明确胃腺癌细胞是否存在瘦素自分泌调节现象和具有合成内源性瘦素的能力。方法采用RT-PCR和组织原位RT-PCR方法检测具有瘦素、瘦素受体蛋白双重表达的胃腺癌组织中瘦素mRNA的表达、细胞定位情况。结果26例具有瘦素和瘦素受体蛋白双重表达的胃腺癌组经RT-PCR和组织原位RT-PCR方法检测发现瘦素mRNA表达均为阳性,定位于胃腺癌细胞的细胞质。结论胃癌细胞自身具有合成和分泌内源性瘦素的功能,存在瘦素自分泌调节现象,可视为胃源性瘦素在胃癌组织中的“异位分泌”。
Objective To study the expression of leptin mRNA and its cellular localization in gastric adenocarcinoma and to clarify whether gastric adenocarcinoma cells regulate autocrine regulation of leptin and the ability to synthesize endogenous leptin. Methods The expression of leptin mRNA and cell localization in gastric adenocarcinoma with double expression of leptin and leptin receptor protein were detected by RT-PCR and in situ RT-PCR. Results Twenty-six cases of gastric adenocarcinoma with double expression of leptin and leptin receptor protein were positive for RT-PCR and in situ RT-PCR. The results showed that the expression of leptin mRNA was positive and located in the cytoplasm of gastric adenocarcinoma cells. Conclusion Gastric cancer cells have the function of synthesizing and secreting endogenous leptin, and there is autocrine regulation of leptin, which can be regarded as the “ectopic secretion” of gastric leptin in gastric cancer.
