目的:研究扩张型心肌病(DCM)大鼠心肌钙调节蛋白和内皮素系统的变化及培多普利和比索洛尔干预的影响。方法:二肾一夹制备高血压糖尿病大鼠模型,部分大鼠用培多普利及比索洛尔治疗,3个月后检测大鼠LV/BW及左室β_1AR、钙调节蛋白、内皮素受体(ETR)等mRNA表达,ETR及亚型B_(max)和K_D值的变化。结果:与正常组相比,DCM大鼠LV/BW增高,SERCA表达下降,活力降低,β_1AR及IP_3-1R上调,ETR数量降低,ET_AR降低为主,培多普利及比索洛尔治疗逆转心室肥厚同时改善钙调节蛋白及内皮素系统的失调。结论:DCM大鼠心室肥厚阶段存在心肌细胞钙调节蛋白及内皮素系统的失调,用培多普利及比索洛尔治疗后可逆转。
AIM: To investigate the changes of cardiac calcineurin and endothelin system in dilated cardiomyopathy (DCM) rats and the effects of perindopril and bisoprolol intervention. Methods: Two rats with one kidney were used to prepare the model of hypertensive diabetic rats. Some rats were treated with pendipril and bisoprolol. After three months, the levels of LV / BW and β_1AR, calcium regulatory protein, endothelin (ETR) and other mRNA expression, ETR and subtypes B_ (max) and K_D value changes. Results: Compared with the normal group, LV / BW increased, SERCA expression decreased, vitality decreased, β_1AR and IP_3-1R increased, the number of ETR decreased, ET_AR decreased, and treatment with trastoprory and bisoprolol reversed the ventricular Hypertrophy improves both dysregulation of calmodulin and endothelin systems. CONCLUSION: Cardiac hypertrophy in DCM rats has dysregulation of cardiomyocyte calmodulin and endothelin system, which may be reversed after treatment with perindopril and bidopril.
