目的 :研究丹参对兔重症急性胰腺炎 (SAP)早期中性粒细胞 (PMN)与内皮细胞 (EC)粘附作用及对血浆肿瘤坏死因子 α(TNF α)的影响。方法 :将日本大耳兔 2 4只建立兔SAP模型 ,随机分为丹参治疗组和对照组。分别于术前 ,术后即刻、2、4、8、16h ,取血分离获得PMN。与体外培养的兔血管EC作用测定PMN EC粘附率 ,并测定各时点血浆TNF α值。结果 :两组术后即刻粘附率均升高 ,对照组术后 2hTNF α开始升高 ,8h达高峰 ,16h仍高于正常 ;治疗组 2、4、8、16h各时点的TNF α的值小于对照组 (P <0 .0 5 )。结论 :丹参能降低SAP血浆TNF α的水平 ,进而抑制PMN EC粘附 ,改善微循环及减轻组织病理损伤
Objective: To study the effect of salvia miltiorrhiza on the adhesion of neutrophils (PMN) and endothelial cells (EC) in rabbits with severe acute pancreatitis (SAP) and to plasma tumor necrosis factor α (TNF α). METHODS: Twenty-four Japanese rabbits were used to establish a rabbit SAP model and randomly divided into Salvia miltiorrhiza treatment group and control group. Preoperatively, immediately after surgery, 2, 4, 8, and 16 h, blood was collected to obtain PMN. The effect of PMN EC adhesion was measured with the rabbit vascular EC cultured in vitro, and the plasma TNFa value was measured at each time point. RESULTS: The adhesion rate of both groups increased immediately after operation. The TNF-alpha level of the control group increased at 2 hours after operation, peaked at 8 hours, and remained higher than normal at 16 hours. TNF-alpha was detected at each time point of 2, 4, 8, and 16 hours in the treatment group. The value is smaller than the control group (P < 0.05). Conclusion :Salvia Miltiorrhiza can reduce the level of TNFα in SAP plasma, inhibit PMN EC adhesion, improve microcirculation and reduce tissue pathological damage
