为检测日本血吸虫小RNA let-7(Sja-let-7)对虫体生殖发育的影响,本研究在日本血吸虫尾蚴感染BALB/c小鼠后的11 d~42 d,将Sja-let-7的同系物(Sja-let-7 mimics)注射入宿主体内对血吸虫Sja-let-7的表达进行干扰,结果显示Sja-let-7 mimics干扰组小鼠肝脏虫卵孵化率显著升高;小鼠肝脏肉芽肿病变减轻十分明显,病灶周围炎性细胞浸润半径变小,空泡变性较对照组轻微,周围正常组织完整性也优于对照组。扫描电镜和透射电镜未观察到虫体的明显变化。利用荧光定量PCR检测Sja-let-7的候选靶基因干扰虫体的表达情况,结果表明Sja-let-7 mimics对大部分候选基因存在不同程度的抑制作用。本研究为后续深入研究小RNA Sja-let-7的功能奠定了基础。
In order to investigate the effect of Sja-let-7 on reproductive development of S. japonicum, we studied the effects of Sja-let-7 on the reproductive development of S. japonicum from 11 d to 42 d after BALB / c mice were infected with Schistosoma japonicum (Sja-let-7 mimics) were injected into the host to interfere with the expression of Sja-let-7 in Schistosoma japonicum. The results showed that the hatching rate of liver in the Sja-let-7 mimics interference group was significantly increased. The lesions of liver granuloma were alleviated obviously. The infiltrative radius of inflammatory cells around the lesion became smaller and the degeneration of vacuoles was less than that of the control group. The surrounding normal tissue integrity was also better than that of the control group. No significant changes of parasites were observed by scanning electron microscopy and transmission electron microscopy. Fluorescent quantitative PCR assay Sja-let-7 candidate target genes interfere with the expression of parasites, the results show that Sja-let-7 mimics have some degree of inhibition of most of the candidate genes. This study lays the foundation for further study on the function of small RNA Sja-let-7.
