探讨补中益气汤对A549/DDP移植瘤裸鼠中Bad,NF-κB,caspase-9,Survivin,m TOR表达的影响。BALB/C裸鼠60只随机分为空白对照组、荷瘤对照组、顺铂组和补中益气汤高剂量+顺铂组(高联组)、补中益气汤中剂量+顺铂组(中联组)、补中益气汤低剂量+顺铂组(低联组),培养A549/DDP细胞(浓度为5×106个/m L),接种于各组,观察各组成瘤情况。给予相应治疗,14 d后取瘤组织固定、切片,免疫组织化学方法、Real-time PCR方法检测各组裸鼠移植瘤中Bad,NF-κB,caspase-9,Survivin,m TOR蛋白和mRNA的表达水平。结果显示补中益气汤联合顺铂可降低移植瘤体积,中联组和高联组的抑瘤率与顺铂组比较,差异有统计学意义(P<0.05);与荷瘤对照组比较,中联组和高联组Bad,NF-κB,Survivin,m TOR的蛋白和mRNA的表达明显减少(P<0.05),caspase-9的蛋白和mRNA在中联组和高联组的表达逐渐增多,与荷瘤对照组比较,差异有统计学意义(P<0.05);Bad,NF-κB,caspase-9的mRNA,中联组和高联组与顺铂组、低联组、荷瘤对照组比较有统计学意义(P<0.05),低联组、顺铂组、荷瘤对照组两两比较,无统计学差异,各因子蛋白和mRNA的表达中联组和高联组比较,无统计学意义。结果表明补中益气汤与顺铂联合作用可抑制A549/DDP移植瘤的生长,补中益气汤抑制A549/DDP移植瘤的作用可能是通过调节Bad,NF-κB,caspase-9,Survivin,m TOR,促进细胞凋亡实现的。
To investigate the effect of Buzhong Yiqi Decoction on the expressions of Bad, NF-κB, caspase-9, Survivin and m TOR in A549 / DDP xenografted nude mice. 60 BALB / C nude mice were randomly divided into blank control group, tumor-bearing control group, cisplatin group and Buzhong Yiqi Decoction high-dose + cisplatin group (high group), Buzhong Yiqi Decoction + cisplatin (Zhonglian group), Buzhong Yiqi Decoction low dose + cisplatin group (low group), A549 / DDP cells (concentration 5 × 106 cells / m L) were inoculated into each group to observe the formation of tumor Happening. The results showed that Buzhong Yiqi Decoction combined with cisplatin can reduce the volume of tumor xenografts, the tumor inhibition rate of Zhonglian group and Guanlian group was significantly lower than that of cisplatin group (P <0.05); Compared with tumor-bearing control group The protein and mRNA expressions of Bad, NF-κB, Survivin and m TOR in Zhonglian group and high-concentration group were significantly decreased (P <0.05). The expressions of caspase-9 protein and mRNA in Zhonglian group and high-linked group were gradually decreased (P <0.05). The mRNA expressions of Bad, NF-κB and caspase-9 in Zhonglian group and Gaolian group were significantly lower than those in cisplatin group, (P <0.05). There was no significant difference in the expression of protein and mRNA between the two groups in the low, the cisplatin and the tumor bearing control groups, No statistical significance. The results showed that Buzhong Yiqi Decoction combined with cisplatin can inhibit the growth of A549 / DDP xenograft tumor. The effect of Buzhong Yiqi Decoction inhibiting A549 / DDP xenograft tumor may be through regulating Bad, NF-κB, caspase-9 and Survivin , m TOR, to promote apoptosis.
