目的 :建立毛细管区带电泳法 (CZE)测定马钱子及其制剂“治伤消淤丸”中士的宁和马钱子碱的含量。方法 :选择含0 .2 m ol/ L醋酸的 2 0 mm ol/ L醋酸铵 (p H 3.6 4)溶液为分离用缓冲液 ,以盐酸多巴胺为内标 ,采用 3.5 k Pa进样 5 s,运行电压2 5 k V,紫外检测波长 2 14nm。结果 :士的宁和马钱子碱的浓度在 5~ 10 0 μg/ ml内时 ,具有良好的线性关系和重现性 ,相关系数 r分别为 0 .9996和 0 .9995 ,RSD<4.5 % ,加样回收率均在 95 %~ 10 5 % ,最低检测限分别为 0 .0 6和 0 .0 4μg/ m l。 结论 :CZE可有效地分离、测定马钱子及其制剂“治伤消淤丸”中的士的宁和马钱子碱的含量 ,方法简便、快速、准确、可靠。
Objective : To establish a capillary zone electrophoresis (CZE) method for the determination of strychnine and strychnine in Zhizhenxiaoxiaoyu Pills. METHODS: A 20 M ol/L ammonium acetate (p H 3.6 4) solution containing 0.2 mol/L acetic acid was used as a buffer for isolation. The dopamine hydrochloride was used as the internal standard and the sample was injected at 3.5 k Pa for 5 s. Operating voltage 25 kV, UV detection wavelength 2 14nm. Results: When the concentration of strychnine and strychnine was within the range of 5 to 10 μg/ml, the linearity and reproducibility were good. The correlation coefficients r were 0 .9996 and 0.995, respectively. The RSD was less than 4.5%. The recoveries of the samples were between 95% and 105%, and the minimum detection limits were 0.06 and 0.04 μg/ml, respectively. Conclusion : CZE can effectively separate and determine the content of strychnine and strychnine in Strychnos and Pills of Zhiqinzi and its preparations. The method is simple, rapid, accurate and reliable.
