利用超临界溶液浸渍法(SSI)将磺胺嘧啶银(AgSD)负载到不对称壳聚糖膜(ACF)制备伤口敷料,研究了SSI过程操作压力及温度对载药量的影响,并对AgSD-ACF的吸水量、水蒸气渗透速率、体外释放、抑菌性能和动物皮肤伤口愈合行为进行了考察.结果表明,AgSD-ACF的载药量随着CO2压力的升高先增加后减少,而温度对载药量的影响较复杂,在20 MPa和50℃时载药量达到最大为5.0 mg/g.AgSD-ACF的水蒸气渗透速率和吸水量可通过改变ACF中致密层的厚度进行调节.AgSD-ACF具有缓释作用,可持续释放10 h以上,并且对金黄色葡萄球菌和绿脓杆菌均有较强的抑制作用.动物试验显示AgSD-ACF伤口敷料具有促进大鼠皮肤伤口愈合的作用.
The wound dressing was prepared by loading silver sulfadiazine (AgSD) onto asymmetric chitosan membrane (ACF) by supercritical solution impregnation (SSI). The effects of operating pressure and temperature on drug loading were studied. The effects of AgSD- ACF water absorption, water vapor transmission rate, in vitro release, antibacterial activity and animal skin wound healing behavior were investigated.The results showed that AgSD-ACF drug loading increased with the increase of CO2 pressure and then decreased, while the temperature The effect on drug loading was complicated, and the maximum drug loading reached 5.0 mg / g at 20 MPa and 50 ° C. The water vapor transmission rate and water absorption of AgSD-ACF could be adjusted by changing the thickness of the densified layer in ACF. AgSD-ACF has a sustained release, sustainable release of more than 10 h, and Staphylococcus aureus and Pseudomonas aeruginosa have a strong inhibitory effect.Animals experiments showed that AgSD-ACF wound dressing has the effect of promoting wound healing in rat skin .
