为研究NaOH处理打破紫椴种子休眠的效果及NaOH处理后对紫椴种子萌发过程中贮藏物质的影响,采用不同质量分数NaOH处理紫椴种子,结合变温层积,定期统计发芽势、发芽率,并在层积过程中定期随机取样进行贮藏物质的测定。结果表明:30%NaOH处理1.5 h对紫椴种子休眠解除效果最好,发芽势平均为38%,发芽率平均为71.66%,20%NaOH处理1.5 h及10%NaOH处理6 h效果次之。与对照相比,NaOH处理后种子在整个层积过程中,淀粉、可溶性蛋白和可溶性糖含量显著下降,淀粉酶活性变化剧烈,呈“W”型。说明NaOH处理能够有效地解除紫椴种子的休眠,同时有效地改善了种子的透水透气性,在一定程度上加速了紫椴种子休眠解除过程中生理生化反应的进程。
In order to study the effect of NaOH treatment on breaking dormancy of Tilia amurense and the effect of NaOH treatment on the storage of Tilia amurensis seed during its germination, different concentrations of NaOH were used to treat Tilia amurensis seed, And in the process of stratification random sampling regularly for the determination of storage material. The results showed that 30% NaOH treatment for 1.5 h had the best effect on seed dormancy release of Tilia amurensis. The average germination potential was 38%, the average germination rate was 71.66%, followed by 20% NaOH for 1.5 h and 10% NaOH for 6 h. Compared with the control, the content of starch, soluble protein and soluble sugar in the seed after NaOH treatment decreased significantly, and the amylase activity changed drastically. The results showed that NaOH treatment could effectively relieve the dormancy of Tilia amurense seedlings and effectively improve the water vapor permeability of seeds, which accelerated the process of physiological and biochemical reactions during the dormancy release of Tilia amurense to some extent.
