目的 了解鼻咽癌肿瘤细胞增殖与凋亡的关系及bcl 2表达在此过程中的作用。方法 6 5例鼻咽癌活检组织及 15例癌旁组织用免疫组化技术检测bcl 2和PCNA ;HE染色中作核分裂像计数 ,并将后两者合并为增殖指数 ;用原位末端标记法 (ISEL法 )显示 44例鼻咽癌及 15例癌旁组织中的细胞凋亡。结果 ①鼻咽癌PCNA、核分裂像数、凋亡细胞数均高于癌旁上皮组织 ,但与组织分型无关 ;增殖指数与凋亡细胞数呈正相关。②bcl 2在鼻咽癌中高表达 ,并与组织分型有关 ,与凋亡细胞数呈负相关。结论 ①鼻咽癌肿瘤细胞具有高水平增殖的同时也具有高水平的凋亡 ,其肿瘤本身是高水平基础上增殖与凋亡失衡的结果。②bcl 2不是调控肿瘤细胞凋亡的唯一因素 ,但其抑制细胞凋亡的功能在鼻咽癌发病机制中有重要作用
Objective To investigate the relationship between the proliferation and apoptosis of nasopharyngeal carcinoma cells and the role of bcl-2 in this process. Methods Sixty-five specimens of nasopharyngeal carcinoma and 15 specimens of para-cancerous tissue were detected by immunohistochemistry for counting bcl-2 and PCNA. The mitotic count was counted by HE staining and the latter two were combined into proliferation index. (ISEL method) showed apoptosis in 44 cases of nasopharyngeal carcinoma and 15 cases of paracancerous tissues. Results ① The numbers of PCNA, mitotic figures and apoptotic cells in nasopharyngeal carcinoma were higher than that in adjacent epithelial tissues, but not with the tissue type. The proliferation index was positively correlated with the number of apoptotic cells. ② The expression of bcl 2 in nasopharyngeal carcinoma was highly correlated with histological type and negatively correlated with the number of apoptotic cells. Conclusions ① The NPC cells have high level of apoptosis as well as high level of apoptosis. The tumor itself is the result of a high level of imbalance between proliferation and apoptosis. ②bcl 2 is not the only factor that regulates tumor cell apoptosis, but its function of inhibiting apoptosis plays an important role in the pathogenesis of nasopharyngeal carcinoma
