目的研究过氧化物酶体增殖物激活受体(Peroxisome Proliferators-activated Receptor,PPAR)-β/δ在兔动脉粥样硬化模型中的表达水平和作用方式。方法 35只雄性新西兰大白兔分为对照组(5只)、高脂组(15只)和球囊损伤组(15只),后两组进一步分为6周、8周、10周组,每组5只。实时定量PCR(Real-time Quantitative PCR)用来检测PPARβ/δm RNA的水平,免疫组织化学法(Immunohistochemistry,IHC)和蛋白质印迹法(Western Blotting,WB)检测PPARβ/δ蛋白的表达,酶联免疫吸附试验(Enzyme-linked Immunosorbent Assay,ELISA)测定兔血清中炎症因子肿瘤坏死因子(tumor necrosis factor,TNF)-α,白介素(interleukin,IL)-6,IL-8和IL-10的水平。结果高脂组和球囊损伤组的PPARβ/δ蛋白质和m RNA水平与对照组相比显著增加(P<0.01)。此外,高脂组PPARβ/δ蛋白质水平在6-10周增加显著(P<0.01),PPARβ/δm RNA在8-10周组之间有显著性差异,6-8周组之间也有明显差异(P<0.05)。PPARβ/δ蛋白质和m RNA水平在球囊损伤组各亚组之间差异显著(P<0.01),而且,球囊损伤组PPARβ/δ的表达水平比同一时间点的高脂组明显增加(P<0.01)。高脂组和球囊损伤组,血清TNF-α、IL-6、IL-8和IL-10的水平与对照组相比明显上升。结论 PPARβ/δ可能参与动脉粥样硬化的进程并发挥重要作用。
Objective To investigate the expression of peroxisome proliferator-activated receptor (PPAR) -β / δ in rabbit models of atherosclerosis and its mechanism of action. Methods 35 male New Zealand white rabbits were divided into control group (n = 5), hyperlipidemia group (n = 15) and balloon injury group (n = 15). The latter two groups were further divided into six weeks, eight weeks and ten weeks Group of five. Real-time quantitative PCR was used to detect the level of PPARβ / δm RNA, the expression of PPARβ / δ protein was detected by Immunohistochemistry (IHC) and Western Blotting (WB) The levels of tumor necrosis factor (TNF) -α, interleukin (IL) -6, IL-8 and IL-10 were determined by enzyme-linked immunosorbent assay (ELISA) Results Compared with the control group, PPARβ / δ protein and mRNA levels in the hyperlipidemic and balloon injured groups were significantly increased (P <0.01). In addition, PPARβ / δ protein levels increased significantly from 6 to 10 weeks in hyperlipidemic group (P <0.01), while PPARβ / δmRNA in 8 to 10-week group were significantly different (P <0.05). The levels of PPARβ / δ protein and m RNA in balloon injury group were significantly different (P <0.01). Moreover, the expression of PPARβ / δ in balloon injury group was significantly higher than that in hyperlipidemia group <0.01). Compared with the control group, the levels of TNF-α, IL-6, IL-8 and IL-10 in the hyperlipidemia group and the balloon injury group were significantly increased. Conclusion PPARβ / δ may participate in the process of atherosclerosis and play an important role.
