背景:应用分子生物学技术与体内/外实验动物模型结合手段,从发育学角度探讨了热休克蛋白(heatshockproteins,HSPs)与胚胎生长发育和畸形发生的内在联系,为充实和推动从分子水平揭示胚胎发育正常或异常的奥秘开辟了一条新的途径。目的:探讨高温、香烟及联合对鼠胚胎脑HSP70及蛋白质合成的影响。设计:随机对照的实验研究。地点、材料和方法:实验在新乡医学院细胞生物学教研室和实验动物中心完成。实验选用SD雄性大鼠100只,雌性大鼠200只合笼过夜,按随机抽签法将150只孕鼠分为高温和高温与香烟联合组,高温组分为37,40,41,42℃4个大组,每一大组随机分为处理时间为1,2,3,4,5min小组,共20组;高温与香烟联合组分为37和42℃两大组,每一大组再分为皮下注射相当于0,0.5,1.0,1.5,2.0支香烟5个小组,共10组。高温组于孕鼠孕第10天进行高温处理,高温与香烟联合组于孕鼠孕第2天进行香烟处理,各组孕鼠高温处理,2h后处死,取胎鼠全脑组织制备匀浆,测量HSP70和蛋白质含量,观察高温及高温+香烟各组胎鼠大脑蛋白质含量及诱导合成的HSP70的含量。主要观察指标:高温、高温与香烟联合对胎鼠脑蛋白质及HSP70变化的影响。结果:高温(40,41,42℃)导致胎脑蛋白质含量从7.01mg下降到5.66mg,与对照组(37℃)比较,差异有显著性意义(F=3.92
BACKGROUND: Molecular biology and in vitro / in vivo experimental animal models are used to explore the intrinsic relationship between heat shock proteins (HSPs) and embryonic growth and development and deformity from the perspective of development. In order to enrich and promote the discovery of molecular levels The mystery of normal or abnormal embryo development opened up a new way. Objective: To investigate the effects of hyperthermia, cigarette and combination on HSP70 and protein synthesis in rat embryonic brain. Design: Randomized controlled experimental study. Location, Materials and Methods: Experiments were performed at Cell Biology Faculty and Laboratory Animal Center of Xinxiang Medical College. 100 SD male rats were selected and 200 female rats were caged overnight. 150 pregnant rats were divided into high temperature and high temperature with cigarette by random sampling method. The high temperature components were 37,40,41,42 ℃ 4 A large group, each group were randomly divided into treatment time of 1,2,3,4,5 min group, a total of 20 groups; high temperature and cigarette joint components of 37 and 42 ℃ two major groups, each sub-group Subcutaneous injection equivalent to 0,0.5,1.0,1.5,2.0 cigarettes 5 groups, a total of 10 groups. In the high temperature group, pregnant rats were exposed to high temperature on the 10th day of pregnancy, and the combined group of high temperature and cigarette was treated with cigarette on the 2nd day of pregnancy. The pregnant rats in each group were subjected to high temperature treatment and sacrificed after 2 hours. The contents of HSP70 and protein were measured. The content of protein and the synthesis of HSP70 in fetal rat brain at high temperature and high temperature + cigarette were observed. MAIN OUTCOME MEASURES: Effects of combination of high temperature, high temperature and cigarette on the changes of protein and HSP70 of fetal rat brain. Results: The protein content of fetal brain decreased from 7.01mg to 5.66mg at high temperature (40,41,42 ℃), which was significantly different from that of control group (F = 3.92
