目的研究钙通道阻滞剂维拉帕米(Verapamil)对人肾癌细胞株GRC-1增殖和凋亡的影响。方法应用四唑盐 (MTT)法和流式细胞仪检测维拉帕米对人肾癌细胞株GRC-1增殖的影响,透射电镜超薄切片观察细胞超微结构检测细胞凋亡。结果MTT比色实验表明维拉帕米50 μmol·L-1、100μmol·L-1对人肾癌细胞株GRC-1增殖的抑制作用明显。流式细胞仪测定细胞增殖周期维拉帕米使大部分细胞停滞于G0/G1期。透射电镜扫描细胞超微结构显示,维拉帕米能促使人肾癌细胞株GRC-1 的细胞凋亡。结论钙通道阻滞剂维拉帕米能抑制人肾癌细胞株GRC-1的增殖,导致细胞凋亡。
Objective To investigate the effect of verapamil, a calcium channel blocker, on the proliferation and apoptosis of human renal cell carcinoma cell line GRC-1. Methods The effects of verapamil on the proliferation of human renal cell carcinoma cell line GRC-1 were detected by MTT method and flow cytometry. The apoptosis of GRC-1 cells was observed by ultrathin section transmission electron microscope. Results MTT colorimetric assay showed that verapamil inhibited the proliferation of human renal cell carcinoma cell line GRC-1 with 50 μmol·L-1 and 100 μmol·L-1. Flow Cytometry to Determine Cell Proliferation Cycle Verapamil caused most of the cells to arrest in the G0 / G1 phase. Transmission electron microscopy of cell ultrastructure showed that verapamil can promote human renal cell carcinoma cell line GRC-1 apoptosis. Conclusion The calcium channel blocker verapamil can inhibit the proliferation of human renal cell carcinoma cell line GRC-1 and lead to apoptosis.
