目的:探讨诱导分化剂联合抗代谢药物对人涎腺腺样囊性癌(SACC)细胞的作用效能,寻找治疗SACC的新途径。方法:应用全反式维甲酸(ATRA)、α-干扰素(α-IFN)和5-氟尿嘧啶(5-FU),分别以单用、双用和联用对涎腺腺样囊性癌-83(SACC-83)细胞株作用,用MTT法、软琼脂集落形成、BrdU掺入法和形态学观察的方法观察细胞的形态和功能变化。结果:①随着单用、双用和联用,SACC细胞的抑制率逐渐升高、平板克隆形成率逐渐下降、DNA合成受抑制。②癌细胞异型性逐渐消失,成熟细胞特征逐渐增多。③药物作用后细胞凋亡现象明显,可见大量凋亡小体。结论:3种药物联合使用时可对SACC细胞产生显著的抑制增殖、诱导分化和促凋亡作用。联合抗代谢药物诱导分化对SACC细胞有明显的抗肿瘤作用。
Objective: To explore the effect of inducing differentiation agent and antimetabolites on human salivary adenoid cystic carcinoma (SACC) cells and to find new ways to treat SACC. Methods: ATRA, α-IFN and 5-fluorouracil (5-FU) were used in the treatment of salivary adenoid cystic carcinoma - 83 (SACC-83) cell lines. Morphological and functional changes were observed by MTT assay, soft agar colony formation assay, BrdU incorporation assay and morphological observation. Results: With the single, double and combination, the inhibition rate of SACC cells gradually increased, the formation rate of plate clone decreased gradually, DNA synthesis was inhibited. ② atypical cancer cells gradually disappear, the characteristics of mature cells gradually increased. ③ After the drug effect of apoptosis was obvious, we can see a large number of apoptotic bodies. Conclusion: The combination of three drugs can significantly inhibit proliferation, induce differentiation and promote apoptosis of SACC cells. The combination of antimetabolite drugs induced differentiation of SACC cells have obvious anti-tumor effect.
