目的探讨库普弗细胞在纤维化肝脏中的表达、分布及意义。方法 Wistar大鼠随机分为健康组、肝纤维化模型组,肝纤维化模型组经皮下注射四氯化碳(CCl4)植物油混合液造模成功。苏木素-伊红染色,肝组织羟脯氨酸(Hyp)含量测定,赖氏法测定丙氨酸转氨酶(ALT),TBA法检测丙二醛(MDA)的水平,鲎试剂法测定血浆内毒素水平,免疫组织化学法测定ED2的表达。结果肝脏纤维化组ALT、MDA、Hyp含量明显高于健康对照组(P<0.05)。ET含量呈上升趋势。免疫组织化学结果显示ED2阳性信号在健康对照组沿肝血窦少量散在分布,纤维化模型组阳性信号明显增多(P<0.05),主要分布在增生的纤维组织及纤维间隔,在肝实质内也有少量分布,阳性信号随着实验的进程呈递增趋势,且阳性细胞胞质丰富、体积变大,外形为星形、多角形和不规则形。结论库普弗细胞活化在肝纤维化发生发展过程中有致关重要的作用。
Objective To investigate the expression, distribution and significance of Kupffer cells in fibrotic liver. Methods Wistar rats were randomly divided into healthy group, liver fibrosis model group and model group of liver fibrosis by subcutaneous injection of carbon tetrachloride (CCl4) vegetable oil mixture success. Hematoxylin-eosin staining, hydroxyproline (Hyp) content in liver tissue, ALT and TBA, and plasma endotoxin level The expression of ED2 was detected by immunohistochemistry. Results The levels of ALT, MDA and Hyp in liver fibrosis group were significantly higher than those in healthy control group (P <0.05). ET content is on the rise. The results of immunohistochemistry showed that there was a small amount of ED2 positive signal in the healthy control group scattered along the hepatic sinusoids, and the positive signals in fibrosis model group were significantly increased (P <0.05), mainly distributed in the proliferated fibrous tissue and fibrous septa, and also within the liver parenchyma A small amount of distribution, the positive signal with the progress of the experiment showed an increasing trend, and positive cells rich in cytoplasm, the volume becomes larger, the shape of stars, polygons and irregular shape. Conclusion Kupffer cell activation plays an important role in the development of liver fibrosis.
