目的:建立以反相高效液相色谱测定葛根中精氨酸含量的方法。方法:以邻苯二甲醛为柱前衍生化试剂;色谱柱为ODS HYPERSIL C_(18)(150mm×4.6mm,5μm),流动相为磷酸二氢钾缓冲液(pH=3.0)-甲醇(65:35),检测波长为340nm。结果:精氨酸进样量在0.1~2.0μg(r=0.999 7)范围内线性关系良好;平均回收率为99.08%,RSD=1.92%(n=6)。结论:本法简便、准确、可靠,适用于葛根中精氨酸的含量测定。
Objective: To establish a method for the determination of arginine in Radix Puerariae by RP-HPLC. Methods: O-phthalaldehyde was used as pre-column derivatization reagent; the chromatographic column was ODS HYPERSIL C18(150mm×4.6mm, 5μm) and the mobile phase was potassium dihydrogen phosphate buffer (pH=3.0)-methanol(65). 35) The detection wavelength is 340 nm. RESULTS: The linearity was good in the range of 0.1-2.0 μg arginine (r=0.999 7). The average recovery was 99.08% and RSD was 1.92% (n=6). Conclusion: This method is simple, accurate, reliable and suitable for determination of arginine in Pueraria lobata.
