尾穗苋凝集素(Amaranthus caudatusagglutin in,ACA)是一种存在于尾穗苋种子中的贮藏蛋白,属苋菜凝集素家族,对同翅目害虫如蚜虫、褐飞虱、叶蝉等有明显的致死作用。植物表达载体pCAMB IA2300-35S-OCS带有35S启动子及终止序列、卡那霉素(Kan)抗性基因NPTⅡ。载体pGEMT-ACA含有ACA基因,通过PCR扩增出ACA基因。把植物表达载体pCAMB IA2300-35S-OCS和扩增后的ACA基因用KpnⅠ和SalⅠ双酶切后经连接酶连接,得到植物表达载体pCAMB IA2300-ACA。将该载体分别导入农杆菌GV3101和LBA4404进行保存。
Amaranthus caudatusagglutin in (Amaranthus caudatusagglutinin, ACA) is a kind of storage protein existing in the Amaranthus retroflexus seeds, which belongs to the amaranth lectin family and has obvious lethal effects on homopteran pests such as aphids, brown planthopper, leafhopper . The plant expression vector pCAMB IA2300-35S-OCS has a 35S promoter and a termination sequence, and a Kan resistance gene NPTII. The vector pGEMT-ACA contains the ACA gene and the ACA gene is amplified by PCR. The plant expression vector pCAMB IA2300-35S-OCS and the amplified ACA gene were double-digested with Kpn I and Sal I and ligated to obtain the plant expression vector pCAMB IA2300-ACA. The vector was introduced into Agrobacterium GV3101 and LBA4404 respectively for storage.
