目的通过观察不对称二甲基精氨酸(ADMA)对单核细胞转化为巨噬细胞过程中分泌单核细胞趋化蛋白-1及植物凝集素样氧化低密度脂蛋白受体-1的影响,探讨ADMA在单核细胞转化为巨噬细胞过程中的作用。方法分别用ADMA、ADMA及L-精氨酸孵育单核细胞48h,酶联免疫吸附法测定上清液中单核细胞趋化蛋白-1的含量,Western blotting法测定细胞中植物凝集素样氧化低密度脂蛋白受体-1的蛋白表达量,逆转录聚合酶链反应法测定单核细胞趋化蛋白-1及植物凝集素样氧化低密度脂蛋白受体-1mRNA的表达。结果 ADMA能够促进单核细胞中单核细胞趋化蛋白-1的分泌(p<0.01),以及上调单核/巨噬细胞中植物凝集素样氧化低密度脂蛋白受体-1蛋白的表达(p<0.05),增加单核/巨噬细胞中单核细胞趋化蛋白-1mRNA(p<0.01)及植物凝集素样氧化低密度脂蛋白受体-1mRNA的表达(p<0.05),而L-精氨酸能抑制ADMA的作用(p<0.05)。结论 ADMA能够增加单核细胞向巨噬细胞转化过程中单核细胞趋化蛋白-1及植物凝集素样氧化低密度脂蛋白受体-1的表达,并且这种作用能被L-精氨酸所抑制。
Objective To investigate the effect of Asymmetric Dimethylarginine (ADMA) on the secretion of monocyte chemoattractant protein-1 and lectin-like oxidized low density lipoprotein receptor-1 during the conversion of monocytes into macrophages , To explore the role of ADMA in the process of monocyte transformation into macrophages. Methods Monocytes were incubated with ADMA, ADMA and L-arginine for 48h respectively. The content of monocyte chemoattractant protein-1 in supernatant was determined by enzyme-linked immunosorbent assay (ELISA), and the lectin-like oxidation LDL receptor-1 protein expression, reverse transcription polymerase chain reaction assay monocyte chemoattractant protein-1 and lectin-like oxidized low density lipoprotein receptor-1mRNA expression. Results ADMA promoted the secretion of monocyte chemotactic protein-1 in monocytes (p <0.01) and up-regulated the expression of plant lectin-like oxidized low density lipoprotein receptor-1 protein in monocytes / macrophages (p <0.01) and Lectin-like Oxidized Low Density Lipoprotein Receptor-1 mRNA (p <0.05) in monocytes / macrophages, while L - Arginine can inhibit the effect of ADMA (p <0.05). Conclusions ADMA can increase the expression of monocyte chemoattractant protein-1 and lectin-like oxidized low density lipoprotein receptor-1 during the transformation of monocytes to macrophages, and this effect can be inhibited by L-arginine Suppressed.
