以玉米叶基部为外植体,诱导出大量愈伤组织,同时对影响愈伤组织诱导和分化的几个重要因素进行探讨。结果表明,不同基因型玉米叶基部愈伤诱导率差异显著,B73愈伤诱导率最高,达74.30%;苗龄为7d时的外植体诱导愈伤效果最理想;2.2mg/L的2,4-D有利于胚性愈伤组织的形成;分化培养基中添加1mg/L 6-BA时铁7922和农大178再生频率最高,添加2mg/L 6-BA时B73和昌7-2再生频率最高。玉米高效再生体系的建立可为进一步开展玉米的遗传转化奠定基础。
With the base of corn leaves as explants, a large amount of callus was induced, and several important factors that affected callus induction and differentiation were also discussed. The results showed that the callus induction rate of maize leaves was significantly different at different genotypes. The callus induction rate of B73 was the highest (74.30%). The callus induction effect of explants at seedling age of 7d was the best. The callus induction rate of 2.2mg / 4-D was beneficial to the formation of embryogenic callus. The regeneration frequency of Tie 7922 and Nongda 178 was highest when 1mg / L 6-BA was added into differentiation medium. When the concentration of 2mg / L 6-BA was added, the regeneration frequency of B73 and Chang 7-2 highest. The establishment of efficient maize regeneration system can lay the foundation for further genetic transformation of maize.
