目的]探讨IL_18及乙肝病毒S基因(HBVS)的联合功效。[方法]从胎肝组织中抽提总RNA ,RT-PCR扩增IL-18cDNA基因。从载体 pcDNA3.0/S中限制性酶切获取HBVS基因 ,然后 ,将其与IL -18cDNA一并亚克隆到真核表达质粒 pIRES-EGFP中。[结果]构建了具有抗肿瘤及乙肝双重功效的IL18_HBVS乙肝基因疫苗。[结论]含有IL_18基因的HBV基因疫苗的构建为基因疫苗的功能和应用研究提供了基础
Objective] To investigate the combined effect of IL_18 and hepatitis B virus S gene (HBVS). [Method] Total RNA was extracted from fetal liver tissue and gene of IL-18 was amplified by RT-PCR. The HBVS gene was obtained by restriction enzyme digestion from the vector pcDNA3.0 / S and then subcloned into the eukaryotic expression plasmid pIRES-EGFP together with the IL-18 cDNA. [Result] The IL18_HBVS Hepatitis B gene vaccine with anti-tumor and hepatitis B dual effects was constructed. [Conclusion] The construction of HBV gene vaccine containing IL_18 gene provides the basis for the function and application of gene vaccine
