目的 应用双向凝胶电泳技术 (two -dimensionalgelelectrophoresis ,2 -DE) ,分离乙肝病毒相关性肝细胞癌 (hepatocellu larcarcinoma ,HCC )的癌组织和癌周组织总蛋白质 ,分析二者之间蛋白质组的差异。方法 以固相pH梯度等电聚焦为第一向 ,垂直板SDS -PAGE为第二向分离肝组织总蛋白质 ,银染显色 ,扫描仪获取凝胶图像并对其进行软件分析。结果 癌组织、癌周组织(肝硬化组织 )、癌周组织 (慢性肝炎组织 )图谱蛋白质点数存在差异 (P <0 0 5 ) ;在与癌组织蛋白质组的差异分析中 ,肝硬化组织和慢性肝炎组织之间存在显著差异 (P <0 0 1)。结论 癌组织与癌周组织的蛋白质组具有差异 ,对这些差异蛋白质点的进一步分析有助于研究HBV相关性HCC的癌变机制及肿瘤标记。
Objective To use two-dimensional gel electrophoresis (2-DE) to isolate hepatocelluar carcinoma (hepatocelluar larcarcinoma, HCC) from cancer tissue and pericancerous tissue and analyze the difference in proteome between them. . Methods The isoelectric focusing of the solid phase gradient was used as the first direction. The SDS-PAGE was used to separate the total protein from the liver tissue in a second direction. The silver staining was used. The gel image was acquired by a scanner and analyzed by software. Results There were differences in the number of protein spots in cancer tissues, pericancerous tissues (cirrhosis tissues), and pericancerous tissues (chronic hepatitis tissues) (P < 0.05); in the analysis of differences between cancer tissues and cancer tissues, cirrhotic tissues and chronic There was a significant difference between the hepatitis tissues (P < 0 01). Conclusions The proteomes of cancer tissues and cancer tissues are different. Further analysis of these differential protein spots can help to study the cancerous mechanism and tumor markers of HBV-related HCC.
