以Au.793菌株为材料,研究了8种营养物质对菌丝生长的影响,并通过正交试验得到深层培养黑木耳菌丝的适宜培养基配方:葡萄糖0.5%,麸皮4%,黄豆粉0.2%。同时确定了深层培养黑木耳液体菌种的工艺条件:温度28℃,培养基pH值6~6.5,罐压0.6kg/cm~2,通气量0.25~0.5v/v/m,搅拌速度500rpm,接种量10~15%,种龄48小时。用液体菌种进行代料栽培,结果表明:液体菌种比传统的固体菌种迟缓期短,污染率低,生长快,可提前10~12天出耳。
Using Au.793 strain as material, the effects of eight nutrients on the mycelium growth were studied. The optimum medium for the further cultivation of Auricularia auricular silkworm was obtained by orthogonal test: glucose 0.5%, bran 4%, soybean flour 0.2%. At the same time, the technical conditions of deep cultivation of black fungus liquid strain were determined: the temperature was 28 ℃, the pH value of the medium was 6 ~ 6.5, the pressure of the tank was 0.6kg / cm ~ 2, the aeration rate was 0.25 ~ 0.5v / Inoculation 10 ~ 15%, seed age 48 hours. Liquid culture of seedlings with liquid culture, the results showed that: liquid bacteria than the traditional solid bacteria slow delay period, low pollution rate, fast growth, 10 to 12 days ahead of the ear.
