目的 研究 2 3 羟基桦木酸对体内、外B16黑色素瘤细胞生长的抑制及诱导凋亡作用。方法 应用MTT分析 ,电镜观察 ,流式细胞仪 ,肿瘤体积大小测定等方法 ,对 2 3 羟基桦木酸诱导黑色素瘤细胞进行检测和观察。结果 2 3 羟基桦木酸体外 3 0~ 80 μg/ml处理 ,体内 3 0 0mg/kg~ 6 0 0mg/kg灌胃时 ,对B16细胞有显著的生长抑制作用 ,有一定的剂量依赖关系并出现大量的凋亡细胞 ,同时体内肿瘤细胞被阻滞在G0 ~G1期 ,DNA合成受阻 ,肿瘤体积缩小。结论 2 3 羟基桦木酸对体内外黑色素瘤细胞增殖有强烈的抑制作用 ,诱导肿瘤细胞死亡的主要途径是凋亡。
Objective To study the inhibitory effect of 2-3 betulinic acid on the growth of B16 melanoma cells in vitro and in vivo and induce apoptosis. Methods MTT assay, electron microscopy, flow cytometry, tumor size determination and other methods were used to detect and observe the melanoma cells induced by 23 betulinic acid. RESULTS: Betella zeae seedlings were treated with 30-80 μg/ml in vitro and 300 mg/kg-600 mg/kg in vivo had significant growth inhibitory effect on B16 cells. There was a dose-dependent relationship between them. A large number of apoptotic cells, while the tumor cells in vivo were arrested in the G0 ~ G1 phase, DNA synthesis was blocked, and the tumor volume was reduced. Conclusion 23 betulinic acid has a strong inhibitory effect on the proliferation of melanoma cells in vitro and in vivo. The main way to induce tumor cell death is apoptosis.
