目的了解糖尿病 (DM)大鼠肾间质转化生长因子 β1 (TGF β1 )表达与纤维粘连蛋白(FN)和胶原Ⅲ型 (ColⅢ )沉积的关系。方法四氧嘧啶复制糖尿病模型 ,免疫组化法检测肾间质TGF β1 、FN和ColⅢ ,PAS染色观察肾组织形态学改变。结果正常肾间质和糖尿病 1周 ,可见FN和ColⅢ ,但未见TGF β1 表达 ,糖尿病 2周开始肾小管见TGF β1 表达 ,并随疾病进展而逐渐增多。糖尿病 1月肾间质中见FN和ColⅢ沉积开始增多 ,2月时更多 ,FN和ColⅢ增多与TGF β1 呈正相关。结论TGF β1促进糖尿病大鼠肾小管间质FN和ColⅢ的沉积。
Objective To investigate the relationship between the expression of transforming growth factor β1 (TGFβ1) and the deposition of fibronectin (FN) and collagen type Ⅲ (Col Ⅲ) in diabetic rats. Methods Alloxan-induced diabetes mellitus model was established. The expression of TGFβ1, FN and Col Ⅲ in renal interstitium was detected by immunohistochemistry. The morphological changes of renal tissue were observed by PAS staining. Results FN and Col Ⅲ were found in normal renal interstitium and diabetes for one week. However, no TGF β1 expression was found in the normal renal interstitium and diabetic rats. TGF β1 expression was observed in the renal tubules 2 weeks after diabetes mellitus, and gradually increased with the progression of the disease. FN and Col Ⅲ deposition increased in renal interstitium in January, more in February, and increased in FN and Col Ⅲ was positively correlated with TGF β1. Conclusion TGF β1 can promote the deposition of FN and Col Ⅲ in the tubulointerstitium of diabetic rats.
