本文采用胰凝乳蛋白酶和胃蛋白酶联合消化方法得到正常人血浆(NHP)有限蛋白酶解产物(NHP-EP)。体外研究发现,NHP的细胞粘附性可达90%;而NHP-EP的细胞粘附抑制性亦可达90%以上,具有可逆性、竞争性、非细胞毒性等特点。动物实验发现,NHP-EP可抑制小鼠胃癌细胞实验性肺转移的形式,抑制率达83.3%;并可延长带瘤小鼠的中位生存期,实验组生存期35天,而对照组为19天。我们认为,NHP-EP对胃癌血行转移防治可能有较大实用价值。
In this paper, the normal human plasma (NHP) limited proteolysis product (NHP-EP) was obtained by the combined digestion of chymotrypsin and pepsin. In vitro study found that NHP cell adhesion up to 90%; and NHP-EP cell adhesion inhibition can also be more than 90%, with reversible, competitive, non-cytotoxicity and other characteristics. Animal experiments have found that NHP-EP can inhibit the form of experimental lung metastasis in mouse gastric cancer cells with an inhibition rate of 83.3%; it can prolong the median survival of tumor-bearing mice. The survival time of the experimental group is 35 days, while the control group is 19 days. We believe that NHP-EP may be of great practical value in prevention and treatment of hematogenous metastasis in gastric cancer.
