利用三种不同溶剂(正己烷、乙醚、氯仿)经索氏提取法提取阿纳其根中的油脂成分,经甲酯化后,用气相色谱质谱法(GC-MS)对脂肪酸组成进行了分析和鉴定。试验结果显示,正己烷提取油脂中共鉴定出了8种脂肪酸,其中亚油酸为36.341%、棕榈酸为34.268%、油酸为19.656%;乙醚提取油脂中共鉴定出了12种脂肪酸。其中亚油酸为34.102%、棕榈酸为32.916%、油酸为20.000%、亚麻酸为5.329%;氯仿提取油脂中共鉴定出了5种脂肪酸。其中亚油酸为42.427%、棕榈酸为40.952%、油酸为11.250%。对阿纳其根脂肪酸进行蛋白酪氨酸磷酸酯酶1B(PTP1B)的抑制作用研究,结果表明脂肪酸对PTP1B有较强的抑制作用,其IC50分别为5.27±0.40、4.71±0.08、7.01±1.98μg/m L。该研究为深入研究阿纳其根的药理作用提供了科学依据。
The constituents of the oil in Anachira root were extracted by Soxhlet extraction using three different solvents (n-hexane, ether and chloroform). After methyl esterification, the fatty acid composition was analyzed by gas chromatography-mass spectrometry (GC-MS) And identification. The results showed that eight kinds of fatty acids were identified in n-hexane oil, including 36.341% of linoleic acid, 34.268% of palmitic acid and 19.656% of oleic acid. Twelve kinds of fatty acids were identified in ethyl ether extract. Among them, 34.102% of linoleic acid, 32.916% of palmitic acid, 20.000% of oleic acid and 5.329% of linolenic acid. Five kinds of fatty acids were identified in chloroform extraction oil. Among them, 42.427% of linoleic acid, 40.952% of palmitic acid and 11.250% of oleic acid. The inhibition of protein tyrosine phosphatase 1B (PTP1B) on the fatty acids of Anatan root showed that the fatty acids had a strong inhibitory effect on PTP1B with IC50 of 5.27 ± 0.40, 4.71 ± 0.08 and 7.01 ± 1.98 μg / m L. This study provides a scientific basis for further study on the pharmacological effects of Anatanine.
