目的:建立一种高效液相色谱法(HPLC)测定儿童血浆中吗替麦考酚酸(MPA)的方法,以进行吗替麦考酚酯的血药浓度监测。方法:采用C18柱(Waters Spherisorb 5μm ODS,4.6 mm×250 mm),流动相:乙腈:水:三乙胺(40:60:0.3)(磷酸调pH至3.5);流速1.0 mL/min;柱温:25℃;检测波长:252 nm。结果:当吗替麦考酚酸浓度在0.5~60mg/L时,其峰面积与浓度的线性关系良好(r=0.996);高、中、低3个浓度的绝对回收率分别为91.5%,93.7%和96.9%,相对回收率分别为103.3%,95.6%和104.7%,日内及日间精密度的RSD均小于5%。目前采用此方法已测定儿童谷浓度血样20份,浓度范围在0.76~2.67 mg/L。结论:本方法测定结果准确、方法简便,可用于儿童患者的血药浓度监测。
OBJECTIVE: To establish a method for the determination of plasma mycophenolic acid (MPA) in children's plasma by high performance liquid chromatography (HPLC), and to monitor plasma concentrations of mycophenolate mofetil. Methods: A C18 column (Waters Spherisorb 5 μm ODS, 4.6 mm × 250 mm) was used. The mobile phase consisted of acetonitrile: water: triethylamine (40:60:0.3) Temperature: 25 ℃; Detection wavelength: 252 nm. Results: When the concentration of mycophenolate mofetil was 0.5 ~ 60mg / L, the linear relationship between peak area and concentration was good (r = 0.996). The absolute recoveries of high, medium and low concentrations were 91.5% 93.7% and 96.9%, respectively. The relative recoveries were 103.3%, 95.6% and 104.7%, respectively. The RSDs of intra- and inter-day precision were less than 5%. Currently used in this method has been determined in children with trough concentrations of 20 blood samples, the concentration range of 0.76 ~ 2.67 mg / L. Conclusion: The method is accurate, the method is simple and can be used for the monitoring of blood concentration in children.
