【目的】探讨过氧化物酶6(PRDX6)在肝脏缺血再灌注损伤模型中的表达及其意义。【方法】建立肝脏缺血再灌注损伤模型,并设立正常对照组、假手术组、缺血再灌注组及槲皮素处理组。分别收集各组肝脏组织并提取总蛋白质,进行双向凝胶电泳、基质辅助激光解析离子化飞行时间质谱鉴定,并用免疫组织化学法对PRDX6的表达进行验证。【结果】双向凝胶电泳显示,缺血再灌注损伤组、槲皮素组分别可检测到大约(825±29)、(831±27)个蛋白质点,经成组比较共计有44个蛋白质点差异明显,上调25个,下调19个。槲皮素能够诱导PRDX6的表达,且在再灌注后12 h点表达最高。【结论】PRDX6可能与槲皮素保护肝脏缺血再灌注损伤有关,将为槲皮素保护肝脏缺血再灌注损伤提供新的实验依据。“,”Objective]To explore the protection mechanism of peroxidase 6 (PRDX6) against liver ische‐mia reperfusion injury .[Methods]A liver ischemia reperfusion injury model was established .And normal con‐trol group ,sham group ,ischemia reperfusion injury group ,and quercetin group were set .Liver tissue was collected from all groups and total proteins were analyzed ,separated by two‐dimensional gel electrophoresis and identified by matrix‐assisted laser desorption ionization time of flight mass spectrometry ,PRDX6 was ver‐ificated by immunohistochemical staining .[Results]Two‐dimensional gel electrophoresis showed that there were about 825 ± 29 protein spots in the ischemia reperfusion injury group ,and 831 ± 27 protein spots in quer‐cetin group .There was a 44 protein spot difference between the two groups ,25 were being up‐regulated ,and 19 being down‐regulated .Experimental results suggest quercetin can induce the expression of PRDX 6 ,and the highest expression was at 12 h after reperfusion .[Conclusion]PRDX6 may be related with protection of quer‐cetin against liver ischemia reperfusion injury ,which is beneficial for the understanding of the mechanism of protection of quercetin against liver ischemia reperfusion injury .
