目的研究一个全新的长链非编码RNA Lnc-CLRN3在肠道天然免疫中的作用。方法应用脂多糖(lipopolysaccharide,LPS)刺激人结肠癌细胞SW480,然后收集细胞提取RNA并通过RT-qPCR筛选表达变化的长链非编码RNA,进一步通过基因过表达、敲除和RNA免疫共沉淀等技术手段对该基因的功能进行深入分析。结果筛选出1个在SW480细胞中高表达且LPS刺激基因表达上调的长链非编码RNA Lnc-CLRN3,该基因主要分布于细胞核内,其转录受到MAPK通路的调控,进一步研究表明该基因通过结合SFPQ蛋白进而影响下游IL12P40等免疫因子的表达。结论长链非编码RNA Lnc-CLRN3参与了肠道细胞的天然免疫调控。
Objective To investigate the role of a novel long-chain non-coding RNA, Lnc-CLRN3, in innate immunity in the intestine. Methods Human colon cancer cell line SW480 was stimulated with lipopolysaccharide (LPS). Then the cells were harvested and RNA was extracted. The long-chain non-coding RNA was screened by RT-qPCR. Overexpression, knockout and coimmunoprecipitation Technical means to conduct an in-depth analysis of the function of the gene. Results A long-chain noncoding RNA, Lnc-CLRN3, which was highly expressed in SW480 cells and upregulated by LPS stimulation, was found. The gene mainly distributed in the nucleus and its transcription was regulated by MAPK pathway. Further studies showed that this gene could bind to SFPQ The protein in turn affects the expression of downstream immune factors such as IL12P40. Conclusion The long-chain non-coding RNA Lnc-CLRN3 is involved in the natural immune regulation of intestinal cells.
