目的研究RhoA和Rock-Ⅱ在Lewis大鼠实验性自身免疫性脑脊髓炎(EAE)模型中的表达,探索其在EAE模型中的作用。方法把36只Lewis大鼠随机分为两组:A组(空白对照组),B组(EAE模型组)。以豚鼠全脊髓为抗原混合福氏完全佐剂(CFA),诱导Lewis大鼠EAE模型,观察实验动物神经功能损害的情况;采用免疫组织化学染色法观察实验动物各组脑、脊髓组织中RhoA、Rock-Ⅱ的表达。结果成功建立Lewis大鼠EAE模型,RhoA和Rock-Ⅱ在EAE模型组的表达较空白对照组明显增加,差异有统计学意义(P<0.01)。结论 RhoA和Rock-Ⅱ在EAE模型中过量表达,可能在EAE模型的发病过程中起关键作用。
Objective To investigate the expression of RhoA and Rock-Ⅱ in Lewis rat model of experimental autoimmune encephalomyelitis (EAE) and explore the role of RhoA and Rock-Ⅱ in EAE model. Methods Thirty-six Lewis rats were randomly divided into two groups: A group (blank control group) and B group (EAE model group). The whole spinal cord of guinea pigs were mixed with Freund’s complete adjuvant (CFA) to induce Lewis rats EAE model to observe the neurological deficits in experimental animals; Immunohistochemical staining was used to observe the expression of RhoA, Rock-Ⅱ expression. Results The Lewis rat model of EAE was successfully established. The expression of RhoA and Rock-Ⅱ in EAE model group was significantly increased compared with the control group (P <0.01). Conclusion Overexpression of RhoA and Rock-Ⅱ in EAE model may play a key role in the pathogenesis of EAE.
