目的探讨增加GSTθ表达水平对人类肝癌7721细胞生长及侵袭能力的影响。方法通过脂质体Lipo-fectAMINE将pcDNA3-GSTθ真核质粒载体转染获得高GSTθ表达水平的7721稳定转染细胞;采用RT-PCR检测GSTθmRNA的表达水平;采用细胞记数法测定细胞的生长;采用Boyden小室方法观察细胞侵袭能力的改变。结果 GSTθ高水平表达对7721细胞的生长无明显影响,但使7721细胞的侵袭能力下降。结论本实验证实了在7721细胞中,GSTθ高水平表达可以降低细胞的侵袭能力。
Objective To investigate the effect of increasing GSTθ expression on the growth and invasion of human hepatocellular carcinoma cell line 7721. Methods Transfection of pcDNA3-GSTθ eukaryotic plasmid vector with Lipo-fectAMINE to obtain 7721 stably transfected cells with high GSTθ expression level. The expression of GSTθmRNA was detected by RT-PCR. Cell growth was measured by cell counting method. Using Boyden chamber method to observe the change of cell invasive ability. Results The high expression of GSTθ had no significant effect on the growth of 7721 cells, but decreased the invasiveness of 7721 cells. Conclusion This experiment confirmed that high expression of GSTθ in 7721 cells can reduce the invasiveness of cells.
