目的:研究杂交瘤细胞的体外高密度培养,大量制备高纯度单克隆抗体(McAb).方法:应用Celi-gen5L细胞培养罐,以半连续灌注方式在1%的低血清条件下培养分泌抗疱疹病毒(HSV)McAb的杂交瘤细胞1A12.结果:细胞密度最高达1.7×107/ml,鼠IgG最高为350μg/ml,连续培养23d,共获鼠IgG5.3g,平均滴度1∶64.结论:半连续式培养杂交瘤细胞,细胞密度和抗体产量均优于纯批式培养.
OBJECTIVE: To study the in vitro high-density culture of hybridoma cells and to prepare high-purity monoclonal antibodies (McAbs) in large quantities. METHODS: Celi-gen5L cells were used to culture 1A12 cells secreting anti-herpesvirus (HSV) McAb under the condition of 1% low serum with semi-continuous perfusion. Results: The highest cell density was 1.7 × 107 / ml and the highest was 350μg / ml. After cultured for 23 days, the mouse IgG5.3g was obtained with the average titer 1:64. Conclusion: The semi-continuous culture of hybridoma cells, cell density and antibody yield were better than the pure batch culture.
