目的:为了制备特殊用途的壳聚糖衍生物质,必须将其相对分子质量控制在4 000~7 000 Da之间,制成窄谱低相对分子质量壳聚糖。方法:采用化学降解方法制成分子质量参差不齐的壳聚糖混合物,然后通过多次分离去除较高及较低分子,最后得到目标产物。结果:采用凝胶过滤色谱(GFC)检测分析,得到相对分子质量4 000~7 000 Da壳聚糖。结论:运用化学降解及多级分离方法可以制备相对分子质量可控的目标分子质量壳聚糖。
OBJECTIVE: In order to prepare special-purpose chitosan derivatives, its relative molecular weight must be controlled between 4,000 and 7,000 Da to prepare narrow-spectrum and low-molecular-weight chitosan. Methods: Chemical degradation method was used to prepare chitosan mixture with different molecular weights. Then the higher and lower molecules were removed by multiple separation to obtain the target product. Results: Gel filtration chromatography (GFC) was used to detect and analyze chitosan with relative molecular mass of 4000-7000 Da. Conclusion: The target molecular weight chitosan with controlled molecular weight can be prepared by chemical degradation and multistage separation.
