目的:研究丙戊酸(VA)药物动力学昼夜节律变化及进食条件对节律的影响.方法:对自由进食及限定进食的ICR小鼠分别以渗透压微泵技术(1.062 mg·h~(-1))及iv(50 mg·kg~(-1))给予丙戊酸钠,并测定VA动力学的时间依赖性变化.结果:血浆VA浓度及清除率在稳态时呈昼夜节律性变化(P<0.01),限定进食时间影响VA动力学的节律,使峰值位相移动约12h.结论:用药时间是影响VA药动学的重要因素.进食条件是VA药动学节律的同步因子之一.
Objective: To study the pharmacokinetics of circadian rhythm of valproic acid (VA) and the effects of eating conditions on rhythm.Methods: ICR mice fed freely and limited intake were treated with osmotic pressure micro-pump (1.062 mg · h ~ (- 1) and iv (50 mg · kg -1) were given sodium valproate, and the time-dependent changes of VA kinetics were determined.Results: The plasma VA concentration and clearance in the steady state showed a circadian rhythm (P <0.01), limiting the feeding time affect the rhythm of VA kinetics, the peak phase shift of about 12h.Conclusion: The medication time is an important factor affecting the pharmacokinetics of VA.Eating conditions is one of the synchronization factors VA pharmacokinetic rhythm .
