目的探讨基质金属蛋白酶9(MMP-9)、细胞周期蛋白依赖性蛋白激酶8(CDK8)和血管内皮生长因子C(VEGF-C)在宫颈癌组织中的表达及临床意义。方法应用二步法免疫组织化学染色对70例宫颈癌组织进行MMP-9、CDK8及VEGF-C蛋白检测,观察三者表达情况及其表达与临床病理特征的关系,并分析宫颈癌组织MMP-9表达与CDK8、VEGF-C表达的关系。结果 MMP-9、CDK8、VEGF-C在临床分期Ⅲ期的阳性表达高于Ⅱ期,有淋巴转移者的阳性表达高于无淋巴转移者,宫颈浸润深度≥2/3者的阳性表达高于深度<2/3者,组织分化低的阳性表达高于高、中分化,差异均具有统计学意义(P<0.05)。宫颈癌组织中MMP-9与CDK8表达呈正相关(rs=0.518,P<0.05),MMP-9与VEGF-C表达呈正相关(rs=0.657,P<0.05)。结论 MMP-9、CDK8和VEGF-C的过表达促进宫颈癌的浸润转移,具有正协同作用。联合检测MMP-9、CDK8和VEGF-C蛋白可作为预测宫颈癌的浸润转移及评价患者预后的生物学指标。
Objective To investigate the expression and clinical significance of matrix metalloproteinase 9 (MMP-9), cyclin-dependent kinase 8 (CDK8) and vascular endothelial growth factor C (VEGF-C) in cervical cancer. Methods The expression of MMP-9, CDK8 and VEGF-C in 70 cases of cervical cancer tissues were detected by two-step immunohistochemical staining. The expression of MMP-9, CDK8 and VEGF- 9 expression and CDK8, VEGF-C expression. Results The positive expression of MMP-9, CDK8 and VEGF-C in stage Ⅲ was higher than that in stage Ⅱ. The positive expression of MMP-9, CDK8 and VEGF-C in stage Ⅲ was higher than those without lymph node metastasis The depth of less than 2/3, low expression of tissue was higher than the high, moderately differentiated, the difference was statistically significant (P <0.05). There was a positive correlation between the expression of MMP-9 and CDK8 in cervical cancer (rs = 0.518, P <0.05), and the positive correlation between MMP-9 and VEGF-C (rs = 0.657, P <0.05). Conclusion The overexpression of MMP-9, CDK8 and VEGF-C promotes the invasion and metastasis of cervical cancer and has a positive synergistic effect. Combined detection of MMP-9, CDK8 and VEGF-C protein can be used as biological indicators to predict the invasion and metastasis of cervical cancer and evaluate the prognosis of patients.
