目的观察内毒素(LPS)致幼年大鼠感染性脑水肿后,腹腔注射孕酮(Prog)对水通道蛋白4(AQP4)表达的影响。方法将90只幼年大鼠随机分为对照组(C,n=10),内毒素组(LPS,n=40)和内毒素+孕酮组(LPS+Prog,n=40)。建立LPS致大鼠感染性脑水肿模型后1h,LPS+Prog组和LPS组分别腹腔注射孕酮和生理盐水,以后每6小时注射一次。于术后6、12、24和48h处死动物,分别行脑组织含水量的测定,用免疫组化和RT-PCR检测脑组织内AQP4表达水平。结果 LPS组脑组织含水量明显高于C组和LPS+Prog组(P<0.01)。在LPS注射6h时,幼鼠脑组织AQP4表达明显增加,12h达高峰。LPS+Prog组在LPS注射后12h时,脑组织中AQP4在蛋白水平和mRNA水平的表达均低于LPS组(P<0.01)。结论孕酮可能在转录和翻译水平下调AQP4的表达,从而抑制LPS诱导的幼年大鼠感染性脑水肿的发生发展。
Objective To observe the effect of progesterone (Prog) on the expression of aquaporin 4 (AQP4) induced by endotoxin (LPS) in young rats. Methods 90 juvenile rats were randomly divided into control group (C, n = 10), endotoxin group (LPS, n = 40) and endotoxin + progesterone group (n = 40). At 1 h after LPS-induced rat model of cerebral edema was established, LPS + Prog and LPS groups were injected intraperitoneally with progesterone and normal saline, respectively, and injected every 6 hours thereafter. Animals were sacrificed at 6, 12, 24 and 48 h after operation. The water content of brain tissue was measured respectively. The expression of AQP4 in brain tissue was detected by immunohistochemistry and RT-PCR. Results The brain water content in LPS group was significantly higher than that in C group and LPS + Prog group (P <0.01). At 6h after LPS injection, the expression of AQP4 in the brain of young rats increased significantly and peaked at 12h. At 12h after LPS + Prog injection, the expression of AQP4 at both protein and mRNA levels was lower in LPS + Prog group than in LPS group (P <0.01). Conclusion Progesterone may down-regulate the expression of AQP4 at the transcriptional and translational levels, thereby inhibiting the development of LPS-induced cerebral edema in young rats.
