目的建立鬼臼毒素醇质体包封率的测定方法。方法采用葡聚糖凝胶微柱离心法分离醇质体与游离药物;用HPLC法测定醇质体中鬼臼毒素,计算包封率。结果在本法选择的色谱条件下,鬼臼毒素得到良好的分离,辅料不干扰测定,鬼臼毒素在2.5~101.1μg/mL(r=0.999 9)线性关系良好。微柱离心法能有效的将鬼臼毒素醇质体与游离药物分离,用该方法测得鬼臼毒素醇质体的包封率为51.03%,RSD为1.24%(n=3)。结论建立了微柱离心-HPLC法,该方法便捷,准确,重现性好,可用于测定鬼臼毒素醇质体的包封率。
Objective To establish a method for determination of entrapment efficiency of podophyllotoxin. Methods The apheresis and free drug were separated by dextran gel micro column centrifugation. The podophyllotoxin was determined by HPLC and the entrapment efficiency was calculated. Results Under the chromatographic conditions selected by this method, podophyllotoxin was well separated and the excipients did not interfere with the determination. The linear relationship between podophyllotoxin and 2.5 ~ 101.1μg / mL (r = 0.999 9) was good. The microcapsule centrifugation can effectively separate the podophyllotoxin ethosome from the free drug. The encapsulation efficiency of podophyllotoxin ethosome was 51.03% and the RSD was 1.24% (n = 3). Conclusion The microcentrifuge-HPLC method was established. The method is convenient, accurate and reproducible. It can be used to determine the entrapment efficiency of podophyllotoxin.
