研究白藜芦醇对镍染毒小鼠胚胎成纤维细胞(NIH/3T3)损伤的保护作用。将NIH/3T3细胞分为镍染毒组和白藜芦醇干预组,染毒组给予终浓度分别为0.00、100.00、200.00、400.00μg/ml的三氧化二镍(Ni_2O_3)混悬液。干预组在染毒前加入白藜芦醇溶液(终浓度为20μmol/L)预处理2 h,其他处理与染毒组一致。采用MTT法检测细胞生长抑制率;实时定量PCR(real time PCR,RT-PCR)法检测细胞中环氧合酶-2(COX-2)mRNA的表达水平。结果显示,随着染毒浓度的升高,染毒组和干预组细胞生长抑制率均明显增加。在Ni_2O_3染毒浓度100.00、200.00、400.00μg/ml时,干预组抑制率均低于染毒组,差异有统计学意义(P<0.05);干预组COX-2mRNA相对表达量分别为4.18±0.18、5.25±1.10、7.34±0.96。干预组COX-2基因相对表达量均低于染毒组,差异有统计学意义(P<0.05)。提示,随着镍染毒浓度的增加,NIH/3T3细胞损伤程度增加,COX-2 mRNA表达水平增高,白藜芦醇对镍所致的细胞损伤具有一定的保护作用。
To study the protective effect of resveratrol on nickel-induced mouse embryonic fibroblasts (NIH / 3T3) injury. NIH / 3T3 cells were divided into nickel exposure group and resveratrol intervention group, and the exposure groups were given nickel oxide (Ni_2O_3) suspension with final concentration of 0.00,100.00,200.00,400.00μg / ml respectively. The intervention group was treated with resveratrol solution (final concentration of 20μmol / L) for 2 h before exposure. The other treatments were the same as those of the control group. The cell growth inhibition rate was detected by MTT assay. The expression of cyclooxygenase-2 (COX-2) mRNA was detected by real-time PCR (RT-PCR) The results showed that with the increase of exposure concentration, the cell growth inhibition rate of the treated group and the intervention group increased obviously. The inhibitory rates of intervention group were lower than that of exposure group when the concentration of Ni_2O_3 was 100.00, 200.00 and 400.00μg / ml (P <0.05), and the relative expression of COX-2 mRNA in intervention group was 4.18 ± 0.18 , 5.25 ± 1.10, 7.34 ± 0.96. The relative expression level of COX-2 gene in intervention group was lower than that in exposure group (P <0.05). These results suggest that the NIH / 3T3 cells have a higher degree of injury and a higher level of COX-2 mRNA expression as the concentration of nickel increases. Resveratrol has a protective effect on nickel-induced cell injury.
