药物致尖端扭转型室速(TdP),为多种药物所引起的少见但又非常严重的副作用,致使多种临床使用有效药物被撤市。因此,探讨和选择有效的临床前实验方法,以预测研发药物发生TdP的倾向性是十分必要的。有学者采用盲法研究提示,兔左心室组织?型模型,在预测药物致TdP发生中具有很高的灵敏度和特异性。这项技术最初用于研究心肌的电异质性和发生QT间期延长及TdP的细胞基础,所应用的评价参数及指标,直接来源于引发TdP的各种关键因素;同时,利用此模型所得出TdP评分方法,还可用于评价候选药物致TdP发生的潜在可能性,以及其与市场上现有同类药物相比所产生TdP的相对危险程度。
Drug-induced torsades de pointes (TdP), a rare but very serious side-effect caused by many drugs, has led to the withdrawal of many clinically useful drugs. Therefore, it is necessary to explore and select effective preclinical experimental methods to predict the propensity of TdP to develop drugs. Some scholars use the blind study suggests that rabbit left ventricular tissue model, in predicting the drug induced TdP with high sensitivity and specificity. This technique was originally used to study the electrical heterogeneity of the myocardium and the QT interval prolongation and the cellular basis of TdP. The evaluation parameters and indices used are directly derived from various key factors that trigger TdP. At the same time, The TdP scoring method can also be used to assess the potential for candidate drug-induced TdP and its relative risk to TdP produced in comparison to currently available medications in the market.
