目的:探讨磁流体高温热疗对小鼠恶性黑色素瘤的治疗作用。方法:接种B16F10黑色素瘤细胞悬液于C57/BL6小鼠的背部皮下,当肿瘤长至直径为(0.8±0.1)cm时,将20mg磁流体直接注射到肿瘤内部,24h后在交变磁场下加温(50℃,10min),间隔24h,连续治疗3次,观察磁流体热疗后肿瘤体积的变化、瘤体的病理变化、对肿瘤生长及小鼠生存期的影响。结果:加温治疗后,部分(4/7)小鼠的瘤体完全消失,实验组肿瘤没有消失的小鼠平均生存(20.333±7.881)d,瘤体消失的小鼠生存>70d;组织学检查肿瘤细胞呈凋亡、坏死样改变。结论:磁流体热疗能有效的抑制小鼠恶性黑色素瘤的生长,延长荷瘤小鼠生存期,为其成为皮肤黑色素瘤的有效治疗手段提供了实验依据。
Objective: To investigate the therapeutic effect of hyperthermia with magnetic fluid on malignant melanoma in mice. Methods: B16F10 melanoma cells were inoculated subcutaneously in the back of C57 / BL6 mice. When the tumor grew to a diameter of (0.8 ± 0.1) cm, 20 mg of magnetic fluid was directly injected into the tumor. After 24 hours, Heating (50 ℃, 10min), 24h intervals, continuous treatment three times, observed changes in tumor volume after magnetic fluid hyperthermia, pathological changes of the tumor, tumor growth and survival of mice. Results: After warming treatment, the tumor completely disappeared in part (4/7) mice, the mice in the experimental group with no disappearance of tumor had an average survival of 20.333 ± 7.881 d, the tumor disappeared in mice survived for> 70 days, Check the tumor cells were apoptotic, necrotic changes. Conclusion: Hydrofluidic hyperthermia can effectively inhibit the growth of malignant melanoma in mice and prolong the survival of tumor-bearing mice, which provides an experimental basis for its application as an effective treatment for cutaneous melanoma.
