目的研究脂质(甘油三酯和极低密度脂蛋白)对大鼠HSC低密度脂蛋白(low-density lipoprotein,LDL)、高密度脂蛋白(hinh-densits lipoprotein, HDL)受体的影响。方法用链霉蛋白酶和胶原酶原位灌流,Nvcodenz密度梯度离心分离大鼠 HSC;应用~(125)I-LDL和~(125)I-HDL_3配体进行放射性配基结合实验测定脂质对 HSC膜 LDL、 HDL受体的影响。结果 HSC膜表面存在 LDL、 HDL受体;脂质可增加 LDL受体对LDL的亲合力,但降低HDL受体对HDL_3的亲合力。结论HSC膜上的LDL、HDL受体对脂蛋白代谢及胆固醇的调节有重要作用,为阐明脂肪肝肝纤维化的发生提供了新的实验依据。
Objective To investigate the effects of lipids (triglyceride and very low density lipoprotein) on low-density lipoprotein (LDL) and high density lipoprotein (HDL) receptors in rat HSC. Methods In situ perfusion with pronase and collagenase and Nccodenz density gradient centrifugation were used to isolate rat HSCs. Radioactive ligand binding assay was performed using ~ (125) I-LDL and ~ (125) I-HDL_3 ligands to determine the effect of lipids on HSC Effect of membrane LDL and HDL receptors. Results There were LDL and HDL receptors on the surface of HSC membrane. Lipid increased the affinity of LDL receptor to LDL but decreased the affinity of HDL receptor to HDL_3. Conclusion The LDL and HDL receptors on HSC membrane play an important role in the metabolism of lipoproteins and the regulation of cholesterol. It provides a new experimental basis for elucidating the occurrence of hepatic fibrosis in fatty liver.
