采用RAPD技术 ,用筛选的 2 2个随机引物对桃属主要种类及其类型进行DNA扩增分析。结果表明 :不同引物扩增出的DNA片段范围在 3~ 13条 ,分子量范围在 30 0bp~ 3kb之间 ,依据特征谱带 ,可建立种间及其类型识别的分子检索表。利用 2 2个引物扩出的 180个位点上的RAPD带 ,进行聚类分析 ,结合前人研究出的结果 ,揭示桃种间亲缘演化顺序为 :内蒙古长柄扁桃→光核桃→山桃→甘肃桃→白花山桃、红花山桃、帚形山桃→桃巴旦、陕西桃巴旦→新疆桃、毛桃。
RAPD technique was used to amplify the major species and their genotypes of peach using 2 2 random primers. The results showed that the DNA fragments amplified by different primers ranged from 3 to 13 and the molecular weight ranged from 30 bp to 3 kb. Based on the characteristic bands, the molecular search table for interspecies and its type identification could be established. RAPD bands at 180 loci amplified by 2 2 primers were used to carry out cluster analysis. Combined with the results of previous studies, the order of phylogenetic evolution of peach cultivars was revealed as follows: Long-stemmed Almonds in Inner Mongolia → Light walnut → Lespedeza → Gansu Peach → white Huashan peach, safflower peach, broom peach → peach Dan, Shaanxi peach Dan → Xinjiang peach, peach.
